Therapeutic and Novel Uses of Adrafinil and Modafinil

Adrafinil is a popular nootropic agent more commonly known for its stimulating properties. This particular cognitive enhancing substance is categorized as a eugeroic agent to which it is able to offer many beneficial effects the user minus the undesirable effects often experienced by individuals using stimulants. Adrafinil is a prodrug of modafinil and both have similar effects. Subjective reports demonstrate that both modafinil and adrafinil increases daytime wakefulness and alertness, motivation, productivity, and mental energy. It also helps in elevating one’s mood and overall energy.

Adrafinil works through various mechanisms to exert its effects. Firstly, it stimulates the alpha-1 adrenergic receptors in your brain. These receptors respond to your noradrenaline (norepinephrine). Note that norepinephrine is linked to concentration, attention, memory, energy and learning. Moreover, it works on your D2 and D3 dopamine receptors. It binds with your dopamine transporters to prevent transmission whilst increase the levels of dopamine in your brain. As a direct result, users report feelings of euphoria, relaxation, lesser anxiety, positivity and overall well-being.

Adrafinil and modafinil in alleviating fatigue

One of the most common symptoms felt by patients diagnosed with some chronic health conditions includes fatigue. The following studies demonstrate how modafinil can help in alleviating fatigue amongst patients suffering from Parkinson’s disease, lung cancer and multiple sclerosis.

Parkinson’s disease

Almost 50% of patients identified from Parkinson’s disease report about suffering from fatigue. Despite this percentage, there remains to be no treatment or PD related fatigue. To assess whether modafinil may help in treating PD related fatigue, 13 patients were given modafinil or placebo for a 4 week treatment. After the study period, data was gathered and analyzed. Data analysis revealed that study participants who were under the modafinil treatment have had decreased fatigue and had their sleep patterns improved as compared to those who took placebo.

Lung cancer

Similar to PD, patients suffering from advanced stage cancer also experiences fatigue. To relieve cancer related exhaustion, healthcare providers always prescribe some CNS stimulants. However, most traditional CNS stimulants have serious adverse effects. Since adrafinil is a well-known stimulating agent that poses little to no minimal risks amongst healthy individuals, modafinil was  administered to patients with non-small cell lung cancer. In this randomized study, participants were fived modafinil doses that would otherwise last for 2 weeks. Data analysis revealed that modafinil was able to overcome fatigue, decrease depression symptoms and reduce excessive daytime sleepiness.

Multiple sclerosis

A total of 50 patients diagnosed with multiple sclerosis were involved in one study. The study participants were treated with low modafinil doses within 3 months. Baseline and subsequent evaluation data demonstrated that improvement was seen amongst 91.5% of the respondents. This particular study only shows that modafinil can definitely help in improving symptoms of fatigue as well as sleepiness.        

Behavior and attention enhancement

Adrafinil was thought to have what they call as behavioral-activating properties. Some studies have shown that modafinil and adrafinil helps in improving mood and vigilance amongst the energy. Both drugs help in improving attention, behavior and mood. Some studies even revealed that modafinil can help in improving attention, depression, sleepiness and psychomotor abilities.

Treatment for narcolepsy

Adrafinil was actually created to induce alertness and wakefulness amongst its users. As such, it is a useful drug for individuals suffering from excessive daytime sleepiness or narcolepsy. To show its efficiency against this particular sleep disorder, study participants were administered with modafinil for 3 weeks. Data analysis revealed that modafinil influenced the activities of the brain’s left hemisphere leading to increased vigilance amongst the study participants.

Another study also revealed similar positive effects. This study involved 151 patients diagnosed with narcolepsy. Participants were given modafinil for 6 weeks. After which, their response to the drug was evaluated. Data evaluation revealed that modafinil can help in improving the quality of life of the participants. It helped patients decrease fatigue and increase cognitive processing, energy and social and physical performance.

Treatment of insomnia

Patients with primary insomnia suffer from daytime sleepiness and fatigue. Since modafinil and adrafinil are known to help induce wakefulness and alertness, some researchers hypothesized that it can also help in decreasing the effect of insomnia in the body. As such, 30 patients were involved in one study. They were randomly involved to three treatments: modafinil only, placebo combined with cognitive behavioral therapy (CBT) and modafinil combined with cognitive behavioral therapy. Data analysis revealed that modafinil combined with CBT treatment is most efficient in improving the sleep patterns of participants. It decreases daytime sleepiness, improve sleep latency and increase compliance to therapy. It was further revealed that modafinil can help in alleviating some of the undesirable effects of CBT.
