What is melanotan I?

Melanotan I, named as Afamelonatide by the developers, is a peptide that was originally created at the University of Arizona. It is a synthetic analog of the naturally produced melanocortin peptide hormone known as alpha-melanocyte stimulating hormone or a-MSH. It has been shown to induce deep tanning or skin pigmentation through the process of melanogenesis. How does it work?

The mechanism of action of melanotan I have been associated to mimicking the natural tanning process. After which, it triggers a series of responses and actions that leads to the production of eumelanin, the photoprotective type of pigment (black or brown) instead of pheomelamin, the non-protective type of pigment (red or yellow).

The earlier binding activity of melanin leads to the stimulation of cyclic adenosine monophosphate (cAMP) production from adenosine triphosphate (ATP) and the activation of AC or adenylate cyclase. In turn, cAMP stimulates protein kinase A (PKA) leading to the synthesis of microphthalmia-associated transcription factor (MITF) protein.

MITF functions through activating specific genes that includes those involved in melanogenesis. This activity then leads to increased levels of melanogenic enzymes in an individual melanocyte. The levels of this enzyme in a melanocyte indicate the cell’s ability to produce eumelanin rather than pheomelanin. The release of eumelanin instead of pheomelanin is the primary mechanism on how melanotan I help in increasing skin pigmentation to provide photoprotection against UV radiations. This is the major therapeutic benefit that can be gleaned from the administration of melanotan I.
